چکیده

Suppressors of the blue fluorescence phenotype of the Arabidopsis trp7-700 mutant can be used to identify mutations in genes involved in plant tryptophan biosynthesis. Two recessive suppressor mutations define a new gene, TRP4. The trp4 mutant and the trp7-700 mutant are morphologically normal and grow without tryptophan, whereas the trp4; trp7700 double mutant requires tryptophan for growth. The trp4; trp7-700 double mutant does not segregate at expected frequencies in genetic crosses because of a female-specific defect in transmission of the double mutant genotype, suggesting a role for the tryptophan pathway in female gametophyte development. Genetic and biochemical evidence shows that trp4 mutants are defective in a gene encoding the fi subunit of anthranilate synthase (AS). Arabidopsis AS fi subunit genes were isolated by complementation of an Escherichia col i anthranilate synthase mutation. The trp4 mutation cosegregates with one of the genes, ASB7, located on chromosome 1. Sequence analysis of the AS67 gene from trp4-7 and trp4-2 plants revealed different single base pair substitutions relative to the wild type. Anthranilate synthase a and p subunit genes are regulated coordinately in response to bacterial pathogen infiltration.

Functional expression of Arabidopsis thaliana anthranilate synthase subunit I in Escherichia coli.

Anthranilate synthase is involved in tryptophan (Trp) biosynthesis. Functional expression of subunit I from Arabidopsis (ASA1) was achieved in bacteria as a protein fused with glutathione S-transferase (GST). The active product was purified in a single step on a glutathione-Sepharose column. The Vmax (45 nmol min-1mg-1), the apparent K(M) for chorismate (180 microM), and the feedback inhibition...

Characterization of rice anthranilate synthase alpha-subunit genes OASA1 and OASA2. Tryptophan accumulation in transgenic rice expressing a feedback-insensitive mutant of OASA1.

Anthranilate synthase (AS) is a key enzyme in the synthesis of tryptophan (Trp), indole-3-acetic acid, and indole alkaloids. Two genes, OASA1 and OASA2, encoding AS alpha-subunits were isolated from a monocotyledonous plant, rice (Oryza sativa cv Nipponbare), and were characterized. A phylogenetic tree of AS alpha-subunits from various species revealed a close evolutionary relationship among OA...

Two anthranilate synthase genes in Arabidopsis: defense-related regulation of the tryptophan pathway.

Arabidopsis thaliana has two genes, ASA1 and ASA2, encoding the alpha subunit of anthranilate synthase, the enzyme catalyzing the first reaction in the tryptophan biosynthetic pathway. As a branchpoint enzyme in aromatic amino acid biosynthesis, anthranilate synthase has an important regulatory role. The sequences of the plant genes are homologous to their microbial counterparts. Both predicted...

Metabolic engineering of the tryptophan and phenylalanine biosynthetic pathways in rice

Aromatic amino acids function as building blocks of proteins and as precursors for secondary metabolism. To obtain plants that accumulate tryptophan (Trp) and phenylalanine (Phe), we modified the biosynthetic pathways for these amino acids in rice and dicot species. By introducing a gene encoding a feedback-insensitive anthranilate synthase (AS) alpha subunit, we successfully obtained transgeni...

Regulation of enzyme synthesis in the tryptophan pathway of Acinetobacter calcoaceticus.

In Acinetobacter calcoaceticus the seven genes coding for the enzymes responsible for tryptophan synthesis map at three chromosomal locations. Two three-gene clusters, one (trpGDC) specifying the small subunit of anthranilate synthase, phosphoribosyl transferase, and indoleglycerol phosphate synthase and the other (trpFBA) specifying phosphoribosyl anthranilate isomerase and both tryptophan syn...
